So having never run NT 3.51 I had to try it. After getting NT 3.51 Workstation installed I found that the sound card had no drivers by default. Following this tutorial: https://www.betaarchive.com/forum/viewtopic.php?t=28315 I got the sound to work in NT 3.51. Steps reproduced here:
Nick007, Betaarchive.com wrote:Open your control panel. For Windows NT 3.51, go to drivers, click add and select the 'creative labs sound blaster 1.X, Pro, 16'. If prompted, insert your Windows installation disc, navigate to D:\i386 (where D is the cdrom drive) and click ok. Leave IQ Adress at 220 and click continue. Now set 'MPU401 IQ address' to disabled. Click OK, Windows wants you to restart, do that. As soon as you logged in again, you should hear, if correct, some sounds.
And then the good ol' fashioned "It is now safe to turn off your computer." That was a while ago...

Do you mean maybe the Extension Pack on the Host? Because if you installed the Guest Additions on the Host, you're doing something wrong... ;)

And there's no "virtual box (6)" BTW, please try to be a little more accurate in your description, there's "VirtualBox 6.0.14" as of this writing...
