Is it possible to use password to protect a VM within a VBox ?
e.g. I can password protect a pdf file.
Can I do the same with VBox

That would have nothing to do with VirtualBox. You would do it from within the OS running in the guest machine.

V5 has disk encryption. You should read up on the downsides before using it: there is no locksmith to call if you lose your key. Even backups will be useless if you forgot what the key was at the time.
