Hi All,
Is there any way we can create a Guest Hard Disk size of 100 GB and with Host free Hard disk size of 50 GB?

Sure, provided it's a dynamic disk. Of course you can't actually store 100GB of data - it will fail when the host runs out of space, and probably lots of stuff on the host will fail too.
In fact, long before the host disk fills up you'll probably start to notice the performance decrease as the host filesystem becomes congested.
Generally speaking, I'm not a big fan of running any resource at or close to 100%.
