Can I relocate the ISO file after the VM is created?

Which ISO are you referring to? If you mean an OS install ISO, that can be discarded once the OS is installed. There's no need to locate it or relocate it.

It's the one that is referred to as "inaccessible" in Virtual Box VM info after I moved it.

Why is it still trying to use the .ISO file? Did you actually install the OS or were you simply running it from the installation .ISO? You need to actually install the OS on the virtual hard disk so that the OS can boot from there.

It is best to unmount an ISO when it's no longer required. To fix your current problem, go to File|Virtual Media Manager, select the CDs tab and release the CD/DVD with the warning tag.
