I see a few references in passing (eg viewtopic.php?f=9&t=18641) to IPv6 being disabled between the guest and host when running with NAT mode. Is that still the case?
And if I used host-only or bridged, is there some way to disable IPv6 for the communication (and have IPv4 only)?

NAT piggybacks on the hosts TCP/IP connection, so if the host doesn't support IPv6 (e.g. XP), then naturally the guest can't use it either.
If you aren't using NAT then the guest is implementing its own independant network stack from top to bottom, so it's entirely up to the guest what network protocols it uses (and AFAIK it needn't even be TCP/IP).

My host(s) are Vista/7/8, which all have IPv6 support. Does that mean IPv6 would be supported? How would I be able to tell?

If I remember correctly IPv6 support is not yet implemented for VBox NAT.

I agree with Martin here. Since the predefined address range is IPv4 I doubt you can force it to use IPv6
You can read 9.12 Fine-tuning the VirtualBox NAT engine in your VBox Manual if you want to try to change it to IPv6 and see.
I also read discussions on this via DEV mail so it may be possible in the future.
