Can guest operate by itself?

Host: Desktop machine with Windows 7 Pro. 64b.

After guest is established, can it be save onto a brand new HDD & operate all by itself?

Are you talking about physical to virtual migration? Your wording makes that seem the case, but I'm not sure. If so...

The VMs hard disk can be imaged like that of any other PC, using a disk imaging tool such as Acronis - ideally with the image saved to a portable (USB) hard drive. The image can be restored onto any other PC, whether physical or virtual. Windows images will very likely need to be reactivated after being moved to a new PC.

Can VirtualBox do the job? No, that is not VBox's purpose. Use a correct tool (such as Acronis as already suggested) and it is easy.
