Just a simple question. Can websites like runescape.com detect if I am using a virtual box or not ? :D

If they use javascript to read the OS hardware, like a video card manufacturer string, then yes, they can. But why would Runescape do that anyway? It's a browser based game that even runs on Macs, so if a system that is not capable of running advanced games, why would they block a VM that might be more capable?
