Do I also have to reinstall the Virtualbox extension after I have updated my 4.0.0 version to 4.0.4 ?

You are not required to do it, but it is advised to avoid possible problems. So far, I haven't heard about them, but better be safe than sorry. It could contain bug fixes and such.
