If there are no snapshots, you can create a new VM and instead of creating a new HDD you attach the original *.vdi file.
You need to try to keep the settings as close as you can to the original.

I had that problem with one vm in 4.0.0 . It was missing from the GUI list. I tried to re-create it using the same .vdi file but got an error that it already existed.
I searched the file system and fould the .vbox file for it and stated it from there. This automatically adds it to the list in the GUI.

Did you make a backup of the .VirtualBox.xml and also the machine xml file?
If you look in the machine xml file you should see the MAC address.
Create a new VM attach the vdi and *BEFORE* you start it edit the MAC information and put in the original.
There really is no other reliable way to install this back with versions previous 4.0.0
