Not in version 3.0.10 you can't, but you will likely be able to in upcoming 3.1.0 version (the beta allows the CD/DVD to be uninstalled/removed).

Oops, didn't look enough, you can add and remove CD/DVD drives

Re: Can I disable the CD/DVD device in Guest?
I've just now added 4 hard disks to a beta VM's IDE controller. (After adding the 4th, it won't let me add any more.)
I created a new VM and booted it off an existing Windows VDI (with the CD/DVD deleted from Settings>Storage) and Windows inside that VM reports NO CD/DVD drive installed (not just an empty drive with no media, but NO drive letter). Device Manager shows NO CD/DVD drive present.
