I use a VBox Guest to download e-mails via Outlook. Is it necessary to install a virus scanner on this guest system or is it sufficient to install a virus scanner only on the host.

If you don't want to infect your Guest, you need one. Each OS is separated and isolated from the other. The Host scanner cannot access the Guest file system.
