Assume I have a linux host with 3 different accounts and only 2 of the 3 have virtual box access.

1) No virtual box access
2) Yes virtual box access
3) Yes virtual box access

Assume I have 2 guest OS. I want user #2 to only access 1 guest OS and user #3 to access both guest OS.

Is this possible?

Yes by using groups and assigning proper rights to the xml folder(s) and VDI location(s).

Host OS access control. It is quite trick to set up especially if your host OS allows multi-user log on. Basically you need to use the VBOX_USER_HOME env variable (see UG) so that the users all point to the same .VirtualBox hierarchy. You can use group membership and group access (or ACLs on Windows) to control if an individual user has access to a given VM. Probably the easiest way is to open everything to the three users apart from the VM directories. Note that if users attempts to access a "forbidden" VM then they error message that they will get will be somewhat confusing for them.
