Setting CD/DVD to .iso

Setting cd/dvd to .iso
should run the games using iso files instead of CD? I can not
it to work.

Care to give us some information other than you can not make it work?

The Microsoft game Age Of Empires calls its CD to work.
I 've created the CD .Iso file and set CD / DVD Rom to point it.
But though Windows see .Iso file in the CD driver it asks to load the AOE2 CD when starts the game ....
Can be played in this way the game ?

If you have the original CD just put it in the cd drive and mount the drive in the guest settings to D:
If you are trying to copy the original cd to use instead of the original (backup) do not burn it as an ISO, just copy it.

I want to play without the CD drive to avoid a break.
Therefore I would like to run the game with the file. Iso

Most likely the game has some kind of protection, preventing you from using the ISO as drive. Something you can try is to install Daemon Tools in the Guest, and put the ISO there. Copy the ISO to the Guest, or share it, and mount it in Daemon Tools.
