I am running an old version (1.6.2) of VirtualBox, on SUSE 10.0. So, I am thinking of upgrading to a more current version.

I have a couple of questions:

* If I upgrade, will VB overwrite my existing VDI definitions? If so, what files do I need to back up?
* Any recommended 2.x versions which have a SUSE 10.0 RPM for easy install? I don't need the very latest version of VB..

Your .vdi files will not be touched. Actually the installation will not touch any of your VM-related files. However, if you start the new VBox version for the first time it will scan all .xml files and most probably updated them to the current version. It will create .bak files. If you consider to go back to 1.6.2 then you need to rename the old files back before you start VBox 1.6.2 again.

Regarding the version: VirtualBox 2.2.4 should be pretty stable. In any case It is advised to use the latest version of a release cycle, that is either 2.0.8, 2.1.4 or 2.2.4.
