I am not able to load an ISO

I am not able to load an ISO, I am using a Vista 64 AMD 64. When I set my DVD players as mouting drive, put in my XP disk I get an error messsage that says:
Cannot open host device 'D"\ for read/write access. Check the permission of that device .
Vbox status code: -38 (VERR_ACCESS_DENIED)
It was working prior to the last update? Any suggestions?

I'm not a Vista user, nor do I often make direct use of host drives, so I'm not certain about this, but the error message you describe seems to indicate that you are trying to read from your hosts CD/DVD drive instead of the ISO image mentioned in your question. It is incorrect to do this even if the ISO image is on a CD/DVD.
While the VM is running you need to go into the "Devices" menu of the guest window. Choose "Mount CD/DVD Rom -> CD/DVD Rom Image...". A dialog box appears, so select your ISO image from the list of those available. If your ISO image is not listed then it means you haven't registered it with VBox yet: so click the "Add" button on the dialog, browse to your ISO, select it and and click ok. That will add it to the list of registered ISOs, select it from that list and it should now be accessible from the guest.
