A few days ago, I installed Android x86 iso into my VBox machine, and unrooted it.
However when I download and click on this app it says that my device is rooted.
I uninstalled and reinstalled the app, as well as rebooted my machine several times but the problem persisted. What else should I do?

I can't say for certain about the rooting, but I do remember having tried an Android x86 VM once, and Whatsapp reported a cutom or modified ROM in the Android OS.
So it is possible that Android x86 does not perform exactly like real Android, and some checks may fail.
FWIW though, it is unlikely that there are any Virtualbox settings that can influence this, unless the host OS's built-in hypervisor being enabled plays a role. If your host Mac is running Ventura or later, the Apple hypervisor is enabled, and cannot be disabled.
You could try running the Android VM on a Linux host, where built-in forced hypervisors aren't a thing. No guarantees, though, whether this will affect the state of rooting inside the VM.

built in hypervisor... how do I check if my mac's hypervisor affected that?

In the VM window's Status Bar, look for the green turtle: If you see it, the host's hypervisor is active.
If instead you see the blue V chip: then full hardware acceleration is available to Virtualbox, and the host hypervisor is disabled.
If you see the green turtle, report what Mac OS you're running and what Virtualbox version you're using, and we'll see if the turtle can be made to swim away.
