Your VMs are not affected if you update VirtualBox. VirtualBox is the program, VMs are the data. If you upgrade iTunes your songs do not get affected. Same with VirtualBox.

Reinstalling Virtual Box didn't resolve the problem. The error code, when trying to start any of my VMs is still the same one I uploaded previously.
Edit:
Upgrading to another Virtual Box Version (5.2.2) resolved the problem for me, maybe the 5.1 version I used before is not compatible with the latest win10 update.

Generally speaking, if you want the latest Windows you should have the latest VirtualBox. You simply didn't mention which VirtualBox version did it for you. Could you?
