My host is windows 7 x64, and I got this error message after uprading VirtualBox-4.2.0-80737-Win.
Failed to create the VirtualBox COM object.
The application will now terminate.

Please let me know how to resolve this problem.

Thanks in advance.

I had this same issue after running windows updates on Win server 2012. I followed some post advice but found that I had to remove, then delete the .VirtualBox on the c:\users\user_name directory. Then reinstall and use my old vdi's and all worked fine. I hope this helps

The issue CO_E_SERVER_EXEC_FAILURE (0x80080005) during GUI startup, can be solved check and change user + system environment variable: VBOX_USER_HOME
In my case it was pointing to an old drive and path which did not exist !
