what is start-up Disk
it show this message and nothing to choose from drop down menu.

When you start the vm it will try to boot from the vms's optical disk. It expects that disc to have a .iso file attached to it containing the installation files for the OS you hope to install.
What OS do you want to install? Where are the installation files for that OS? If they are an ISO file on your host, assign that file to the optical disc of the vm. If they are on a CD/DVD, put that disc in the host's drive and select the option to use the host's drive.
To assign the file or the disc to the optical drive, go to the Settings of the vm and select storage. Click the optical drive (which will be showing Empty) then click the blue disk at the right (see screenshot). Select from the dropdown list.

FWIW, the dropdown list shows only the ISO files already known to VirtualBox. Use the button right next to it to select a new ISO file.
