VB fails to run on Preview Build 14316
Virtualbox installs but fails to run on the latest build 14361 as well. Interesting the program does not show up in Add/Remove programs but can be uninstalling using the Iobit Uninstaller. Seems like all the directories and registry entries are there and can be removed by the uninstaller yet the programs dies without a peep when you try to run it.

Re: VB fails to run on Preview Build 14316
I am not having this issue.
When the issue happens, as it crashes, do you see a "WerFault.exe" process run in Task Manager briefly? Also, can you see if any events are generated in Event Viewer, whenever it fails to start?
Honestly, this sounds like something specific to your system or setup, rather than a specific VirtualBox issue. Not sure.

Re: VB fails to run on Preview Build 14316
You didn't ask me for more details, but I thought I would answer. This is what I see in the Event Viewer under Windows Logs -> Application
Also see attachment with a log that I retrieved from the event viewer.
I briefly see Windows Problem Reporting showing up in my Task Manager, so I opened it up and I saw a log for each time I started VB. Here is an example of what it reports:
I hope this helps

crashingdutchman:
Your VirtualBox startup issue is the exact same issue that was widely reported in this thread:
viewtopic.php?f=38&t=76813
It has already been resolved, as of 4/4/2016, per the following posts:
viewtopic.php?f=38&t=76813&start=15#p358641
viewtopic.php?f=38&t=76813&start=30#p358671
viewtopic.php?f=38&t=76813&start=30#p358944
The latest VirtualBox Test Builds are available here:

mfelker: Does this also fix the startup issue you were having?
Good luck!
Jacob

I had the same problem. The test build seems to run fine. Thanks.
Re: VB fails to run on Preview Build 14316
Just wanted to note here that this (the preview version) also worked for me on Windows 10 Build 14316 as well.
EDIT: To update, although the manager starts up, every time I try to boot a Windows 7 guest on the box (that worked before upgrading Windows 10 and VB) - it gives me a BSOD, System Stop Exception (if i remember correctly). If there's anything I can provide to help - let me know

Re: VB fails to run on Preview Build 14316
Strange as i got it working in 2 different Vm's using 5.0.16 r105871
Re: VB fails to run on Preview Build 14316
It also fails on the latest Preview build 14328.rs1_release.160418-1609
I installed the latest test build Virtualbox-5.0.19-106806-win.exe
This is what I see in the event viewer:

Re: VB fails to run on Preview Build 14316
guivho:
The startup problem with Windows 10 Insider Build 14328, while looking similar to prior problems, is actually a different problem than the one in this thread.
So ... let's consolidate efforts, here in the proper "14328" thread for the new problem:

Thanks,
Jacob
