I want to make sure that my users don't violate the Apple EULA by accident. If I allow my users to create and run any VM they want, is there a mechanism in place that blocks the install of MacOS X Client version? Like to detect the disc and not allow it to boot?

There are a few things that prevent them from installing OSX. A genuine OS X installer will fail when it's not run on a genuine Mac. In addition, VB will show strange behaviour like guru meditation and crashes. If you want to be sure that they don't install it, instruct them to not do it in the first place. Have some system in place that keep them in check (will probably be not computer regulated).
